Immune synapses: mitochondrial morphology matters.

چکیده
Proper positioning of mitochondria is critical for cellular function. Mitochondria localization close to synapses regulates signalling at neuronal and immune synapses (ISs). Vice versa, synapses influence activity, motility and the fusion/fission balance of close-by mitochondria. In this issue of The EMBO Journal, Baixauli et al (2011) identify a role for the mitochondrial fission factor dynamin-related protein 1 (DRP1) at the IS. DRP1 not only controls mitochondrial fission but also mitochondrial positioning to the IS, thereby modulating IS formation and downstream signalling. Main functions of mitochondria are energy supply, Ca buffering, supply of metabolites and the sequestration of apoptotic factors (Hollenbeck and Saxton, 2005; Chan, 2006). It is not surprising that mitochondria thereby regulate a majority of cellular processes. In a simple model,

Sideroflexin 3 is an α-synuclein-dependent mitochondrial protein that regulates synaptic morphology
α-Synuclein plays a central role in Parkinson's disease, where it contributes to the vulnerability of synapses to degeneration. However, the downstream mechanisms through which α-synuclein controls synaptic stability and degeneration are not fully understood. Here, comparative proteomics on synapses isolated from α-synuclein-/- mouse brain identified mitochondrial proteins as primary targets of...
متن کاملMitochondrial trafficking to synapses in cultured primary cortical neurons.
Functional synapses require mitochondria to supply ATP and regulate local [Ca2+]i for neurotransmission. Mitochondria are thought to be transported to specific cellular regions of increased need such as synapses. However, little is known about how this occurs, including the spatiotemporal distribution of mitochondria relative to presynaptic and postsynaptic sites, whether mitochondria are dynam...
متن کاملTime for integration: communication in the immune system.
Signals exchanged between cells in the immune system are generated at specific contact regions termed "immunological synapses" (IS). The morphology and the functions of IS vary with circumstances and include directing secretion and integrating positive and negative signals to tune the extent of response - a model that is essentially derived from neurosciences. Indeed, the mechanisms of signal i...
متن کاملMitochondria, Oxidative Stress, and Pollutants including Thimerosal
A common feature in the early stages of many neurodegenerative diseases lies in mitochondrial dysfunction, oxidative stress, and reduced levels of synaptic transmission. Many genes associated with neurodegenerative diseases are now known to regulate either mitochondrial function, redox state, or the exocytosis of neurotransmitters. Mitochondria are the primary source of reactive oxygen species ...
متن کاملKupfer-type immunological synapse characteristics do not predict anti-brain tumor cytolytic T-cell function in vivo.
To analyze the in vivo structure of antigen-specific immunological synapses during an effective immune response, we established brain tumors expressing the surrogate tumor antigen ovalbumin and labeled antigen-specific anti-glioma T cells using specific tetramers. Using these techniques, we determined that a significant number of antigen-specific T cells were localized to the brain tumor and su...
